      <title>Introduction</title>
<p>Earthworms play an important role in soil ecosystems, contributing significantly to organic matter decomposition and nutrient cycling (<xref ref-type="bibr" rid="B10">Edwards et al. 2022</xref>; <xref ref-type="bibr" rid="B27">Li et al. 2026</xref>). Among the diverse earthworm taxa, <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Amynthas">Amynthas</tp:taxon-name-part></tp:taxon-name> Kinberg, 1867 belongs to <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="family" reg="Megascolecidae">Megascolecidae</tp:taxon-name-part></tp:taxon-name> Rosa, 1891 and is a typical earthworm of the Oriental and Palearctic realms (<xref ref-type="bibr" rid="B48">Xu 1996</xref>). It is also the most widely distributed and species-rich genus in China (<xref ref-type="bibr" rid="B21">Jiang and Qiu 2018</xref>; <xref ref-type="bibr" rid="B17">Han et al. 2024</xref>, <xref ref-type="bibr" rid="B18">2026</xref>). The classification system established by <xref ref-type="bibr" rid="B40">Sims and Easton (1972)</xref>, centered on the position and number of spermathecal pores, has provided an important framework for species delimitation within <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Amynthas">Amynthas</tp:taxon-name-part></tp:taxon-name>. Within this system, the <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Amynthas">A.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="sieboldi">sieboldi</tp:taxon-name-part></tp:taxon-name> group is defined by three pairs of spermathecal pores located in 6/7–8/9. However, the phylogenetic relationships within this species group have not been fully tested.</p>
      <p>COI barcoding combined with Assemble Species by Automatic Partitioning (<abbrev xlink:title="Assemble Species by Automatic Partitioning">ASAP</abbrev>) (<xref ref-type="bibr" rid="B34">Puillandre et al. 2021</xref>) and Generalized Mixed Yule Coalescent (<abbrev xlink:title="Generalized Mixed Yule Coalescent">GMYC</abbrev>) (<xref ref-type="bibr" rid="B32">Nee et al. 1994</xref>) species delimitation methods has been widely used for earthworm species identification (<xref ref-type="bibr" rid="B13">Fernández et al. 2012</xref>; <xref ref-type="bibr" rid="B33">Novo et al. 2012</xref>; <xref ref-type="bibr" rid="B16">Goulpeau et al. 2022</xref>; <xref ref-type="bibr" rid="B28">Liu et al. 2025a</xref>, <xref ref-type="bibr" rid="B29">2025b</xref>, <xref ref-type="bibr" rid="B30">2026</xref>; <xref ref-type="bibr" rid="B35">Qiao et al. 2026</xref>). However, the standard COI barcode, with a fragment length of merely 658 bp, exhibits limited resolution in resolving specific phylogenetic relationships. Consequently, <xref ref-type="bibr" rid="B2">Chang and James (2011)</xref> advocated the incorporation of longer sequences derived from both mitochondrial and nuclear genomes. Accordingly, concatenated mitogenomic protein-coding genes (<abbrev xlink:title="protein-coding genes">PCGs</abbrev>) exceeding 10 kb in total length have emerged as a robust data for reconstructing high-resolution phylogenies of earthworms (<xref ref-type="bibr" rid="B55">Zhao et al. 2022</xref>, <xref ref-type="bibr" rid="B56">2025</xref>; <xref ref-type="bibr" rid="B38">Sato et al. 2023</xref>; <xref ref-type="bibr" rid="B1">Aspe et al. 2025</xref>; <xref ref-type="bibr" rid="B18">Han et al. 2026</xref>; <xref ref-type="bibr" rid="B47">Xu et al. 2026</xref>).</p>
      <p>The megascolecid earthworm fauna of northwestern China (Gansu and Shaanxi provinces) remains poorly investigated (<xref ref-type="bibr" rid="B15">Gates 1935</xref>; <xref ref-type="bibr" rid="B6">Chen et al. 1975</xref>, <xref ref-type="bibr" rid="B3">1995</xref>; <xref ref-type="bibr" rid="B11">Feng 1984</xref>; <xref ref-type="bibr" rid="B12">Feng and Ma 1987</xref>). Integrating morphological examination, COI barcode-based species delimitation, and mitogenomic phylogenetic analysis, the present study aims to: (1) describe two new taxa, <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Amynthas">Amynthas</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="longnanensis">longnanensis</tp:taxon-name-part></tp:taxon-name> sp. nov. and <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Amynthas">A.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="pectiniferus">pectiniferus</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="subspecies" reg="guanzhongensis">guanzhongensis</tp:taxon-name-part></tp:taxon-name> ssp. nov., both belonging to the <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Amynthas">A.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="sieboldi">sieboldi</tp:taxon-name-part></tp:taxon-name> group and distributed in northwestern China; and (2) elucidate their phylogenetic positions within this group.</p>

The two taxa are thus separated by a substantial geographic distance.</p>
<p>The Pleistocene epoch (ca. 2.58–0.0117 million years ago) was marked by recurrent alternations between glacial and interglacial periods (<xref ref-type="bibr" rid="B7">Crucifix 2012</xref>), during which climate oscillated between cold–arid and warm–humid conditions. In the East Asian monsoon domain, interglacial phases were characterized by intensified monsoons delivering warm, humid climates, whereas glacial periods featured weakened monsoons and cold, arid conditions (<xref ref-type="bibr" rid="B50">Yang et al. 2019</xref>). Megascolecids are strictly moisture-dependent and highly sensitive to desiccation (<xref ref-type="bibr" rid="B22">Kaneda 2023</xref>). Accordingly, humid interglacial intervals likely provided dispersal corridors connecting suitable habitats, thereby facilitating population migration. Conversely, arid glacial periods rendered lowland plains inhospitable, obstructing gene flow and fostering geographic isolation. These recurrent Pleistocene climatic oscillations thus triggered successive cycles of dispersal, isolation, and differentiation. The disjunct distribution pattern—<tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Amynthas">A.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="pectiniferus">pectiniferus</tp:taxon-name-part></tp:taxon-name> being widespread across southeastern China versus <tp:taxon-name><tp:taxon-name-part taxon-name-part-type="genus" reg="Amynthas">A.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="species" reg="pectiniferus">p.</tp:taxon-name-part> <tp:taxon-name-part taxon-name-part-type="subspecies" reg="guanzhongensis">guanzhongensis</tp:taxon-name-part></tp:taxon-name> ssp. nov. being restricted to Weinan—likely reflects divergence driven by Pleistocene glacial–interglacial climatic fluctuations.
